Job Information

University of Rochester Technical Assoc I (S) in Rochester, New York

POSITION SUMMARY:

The Hematology and Transplantation laboratories conduct basic research as well as translational investigations. The individual occupying this position will be a critical component of the laboratory with responsibility for developing and implementing cellular and molecular biology experiments that will be utilized in investigating interactions between primary cells from leukemia or hematopoietic stem cells with components of the marrow microenvironment in 3D-models. We are focused on examining the metabolic changes as well as molecular changes in our culture model. These techniques will include cell viability/toxicity assays, flow cytometry, PCR, Western blots, histology, immunocytochemistry, live imaging, and ELISA. This individual will also be responsible for conducting animal studies in the laboratory. These studies include engraftment of hematopoietic stem cells or leukemia cells within immune suppressed mice after sub-lethal irradiation. These procedures will include but are not limited to irradiation of animals with cesium source, tail vein injections, monitoring animals, obtaining blood by submandibular blood collecting technique, sacrificing and harvesting organs and performing flow cytometry on tissue and cells. In addition, the technical associate will help with flow cytometry and ELISA studies done as part of clinical trials.

SUPERVISION :

This position reports to Dr. Omar Aljitawi and requires that the individual will be able to use independent judgment in planning of experiments, as well as the ability to troubleshoot and modify methods as required.

TYPICAL ASSIGNMENTS AND RESPONSIBILITIES :

To provide research support to the principal investigator by independently coordinating and conducting the execution of experiments to better understand the function and biological properties of hematopoietic and leukemic cells as well as immunologic responses in transplant recipients. Experimental design will be determined with input from the laboratory investigator and other project collaborators.

MOLECULAR BIOLOGY TECHNIQUES

15% Enhance the molecular biology capabilities of the laboratory by independently designing and performing research experiments utilizing complex molecular biology techniques including isolation of RNA and DNA, in situ hybridization, RT-PCR, and qRT-PCR, western blots, and microarrays.

CELL BIOLOGY

15% Compensate and analyze imaging flow cytometry data; independently perform flow cytometry analysis and present data to PI and other collaborators.

10% Prepare cells by staining for flow cytometry analysis, multi-parameter flow cytometry, cell sorting.

10% Independently perform various cell culture and colony forming assays as well as manipulation of murine and human cell lines and primary cultures for in vivo and in vitro analysis

ANIMAL PROTOCOLS

10% Responsible for designing animal studies using various agents to study engraftment and migration of human cells over time by flow cytometric assays.

5% Harvesting and processing of tissues from experimental and control animals

5% Perform whole body sublethal irradiation experiments and make decisions regarding animal experiments.

LABORATORY RESPONSIBILITIES

5% Advise principal investigator regarding scientific techniques that are to be employed and make suggestions for design development and analysis of research projects.

5% Present data at lab meetings, floor meetings, and conferences. Collaborate with other laboratory members and collaborators. Provide oral and written summaries, develop figures for publications, and assist with the preparation of research papers and grants.

5% Participate with on-going clinical trial experiments such as colony proliferation, apoptotic studies and other correlative studies including flow cytometry and ELISA.

5% Contribute to routine laboratory maintenance and orderliness. Contribute to oversight of new laboratory members and in the ordering of supplies. Oversee utilization of various instrumentation and lab equipment.

REQUIREMENTS:

Bachelor's degree with major course work in field of assignment, and 1 year of related work experience involving direct assistance to researchers; or any equivalent combination of experience, training and education.

Basic laboratory experience is required. Familiarity with whole body irradiation, multispectral imaging flow cytometry, FACS, cytometry analysis software, and molecular biology techniques desired. Excellent oral and written communications skills and ability to work in a collaborative work environment. Candidate must be able to work with rodents and must complete human subjects research training.
